The SkyBus runs to Southern Cross Station. From there, Melbourne's free CBD tram zone covers most central hotels; if your hotel is outside the free zone you will need a Myki fare or a short taxi ride on top, both charged by their own operators.
The real cost of SkyBus is time, not money. The bus-to-station-to-tram-to-hotel chain can add 30-45 minutes compared to a direct transfer, especially if the tram timing does not line up.

Family of two adults and two children: SkyBus works out at AUD 65.00 one way. One vehicle door-to-door avoids the bus-tram-walk logistics with tired kids, and it is worth pricing properly rather than assuming the bus wins.
Group of five or six: per person, one van is usually the best value for door-to-door service, because SkyBus charges each head while a vehicle charges once.
